چکیده

70 A large percentage of the topics covered at the 10th Conference on Retroviruses and Opportunistic Infections in Boston were concerned with basic research aspects of HIV-1 and AIDS. Topics that generated particular interest included the identification of host factors that restrict retrovirus and lentivirus infection. In addition, a number of presentations concerned the use of RNA interference (RNAi) as a tool for understanding HIV biology, and several presentations discussed approaches to therapeutic exploitation of RNAi in HIV/AIDS.

Clearance of a productive lentivirus infection in calves experimentally inoculated with caprine arthritis-encephalitis virus.

Lentiviruses have long been considered host-specific pathogens, but several recent observations demonstrated their capacity to conquer new hosts from different species, genera, and families. From these cross-species infections emerged new animal and human infectious diseases. The successful colonization and adaptation of a lentivirus to a nonnatural host depends on unspecific and specific host ...

Plastic Proteins and Monkey Blocks: How Lentiviruses Evolved to Replicate in the Presence of Primate Restriction Factors

Restriction factors are cellular factors that block virus infection, either by direct interaction with viral factors or by rendering the cellular environment incompatible with viral replication. Wellcharacterized restriction factors include SAMHD1 [1,2], BST-2/ tetherin [3,4], APOBEC3G [5], and TRIM5a [6]. In general, viruses evolve resistance to the restriction factors of their natural hosts b...

Primate lentivirus capsid sensitivity to TRIM5 proteins.

Mammalian cells express several factors that inhibit lentiviral infection and that have been under strong selective pressure. One of these factors, TRIM5, targets the capsid protein of incoming retrovirus particles and inhibits subsequent steps of the replication cycle. By substituting human immunodeficiency virus type 1 capsid, we were able to show that a set of divergent primate lentivirus ca...

Capsid-dependent and -independent postentry restriction of primate lentivirus tropism in rodent cells.

Retrovirus tropism can be restricted by cellular factors such as Fv1, Ref1, and Lv1 that inhibit infection by targeting the incoming viral capsid. Here, we show that rodent cells exhibit differential sensitivity to infection by vesicular stomatitis virus G-pseudotyped lentiviruses and that differences between human immunodeficiency virus type 1 and simian immunodeficiency virus (SIVmac) infecti...

Cellular Restriction Factors of Feline Immunodeficiency Virus

Lentiviruses are known for their narrow cell- and species-tropisms, which are determined by cellular proteins whose absence or presence either support viral replication (dependency factors, cofactors) or inhibit viral replication (restriction factors). Similar to Human immunodeficiency virus type 1 (HIV-1), the cat lentivirus Feline immunodeficiency virus (FIV) is sensitive to recently discover...
